Metabolite Identification
Common NameN-Desmethyltamoxifen
DescriptionN-Desmethyltamoxifen belongs to the class of organic compounds known as stilbenes. These are organic compounds containing a 1,2-diphenylethylene moiety. Stilbenes (C6-C2-C6 ) are derived from the common phenylpropene (C6-C3) skeleton building block. The introduction of one or more hydroxyl groups to a phenyl ring lead to stilbenoids. N-Desmethyltamoxifen is a very strong basic compound (based on its pKa). Within humans, N-desmethyltamoxifen participates in a number of enzymatic reactions. In particular, N-desmethyltamoxifen and formaldehyde can be biosynthesized from tamoxifen; which is mediated by the enzymes cytochrome P450 3A4 and cytochrome P450 3A5. In addition, N-desmethyltamoxifen can be converted into endoxifen; which is catalyzed by the enzyme cytochrome P450 2D6. In humans, N-desmethyltamoxifen is involved in tamoxifen metabolism pathway. The introduction of one or more hydroxyl groups to a phenyl ring lead to stilbenoids. N-Desmethyltamoxifen is a metabolite of Tamoxifen. N-Desmethyltamoxifen is only found in individuals that have used or taken Tamoxifen. Stilbenes (C6-C2-C6 ) are derived from the common phenylpropene (C6-C3) skeleton building block. These are organic compounds containing a 1,2-diphenylethylene moiety.

Enzymes

General function:
Involved in flavin-containing monooxygenase activity
Specific function:
Involved in the oxidative metabolism of a variety of xenobiotics such as drugs and pesticides. It N-oxygenates primary aliphatic alkylamines as well as secondary and tertiary amines. Plays an important role in the metabolism of trimethylamine (TMA), via the production of TMA N-oxide (TMAO). Is also able to perform S-oxidation when acting on sulfide compounds.

General function:
Involved in sequence-specific DNA binding transcription factor activity
Specific function:
Nuclear hormone receptor. Binds estrogens with an affinity similar to that of ESR1, and activates expression of reporter genes containing estrogen response elements (ERE) in an estrogen-dependent manner. Isoform beta-cx lacks ligand binding ability and has no or only very low ere binding activity resulting in the loss of ligand-dependent transactivation ability. DNA- binding by ESR1 and ESR2 is rapidly lost at 37 degrees Celsius in the absence of ligand while in the presence of 17 beta-estradiol and 4-hydroxy-tamoxifen loss in DNA-binding at elevated temperature is more gradual

General function:
Involved in ATP binding
Specific function:
Xenobiotic transporter that may play an important role in the exclusion of xenobiotics from the brain. May be involved in brain-to-blood efflux. Appears to play a major role in the multidrug resistance phenotype of several cancer cell lines. When overexpressed, the transfected cells become resistant to mitoxantrone, daunorubicin and doxorubicin, display diminished intracellular accumulation of daunorubicin, and manifest an ATP- dependent increase in the efflux of rhodamine 123
