Metabolite Identification
Common NameTioconazole
DescriptionTioconazole, also known as vagistat-1 or gyno-trosyd, belongs to the class of organic compounds known as benzylethers. These are aromatic ethers with the general formula ROCR' (R = alkyl, aryl; R'=benzene). They are available in one day doses, as opposed to the 7-day treatments more common in use in the past. Tioconazole is an antifungal medication of the imidazole class used to treat infections caused by a fungus or yeast. Tioconazole topical (skin) preparations are also available for ringworm, jock itch, athlete's foot, and tinea versicolor or "sun fungus". These side effects may be only temporary, and do not normally interfere with the patient's comfort enough to outweigh the end result. Tioconazole is a drug which is used for the local treatment of vulvovaginal candidiasis (moniliasis). Side effects (for the women's formulas) may include temporary burning/irritation of the vaginal area, moderate drowsiness, headache similar to a sinus headache, hives, and upper respiratory infection. Tioconazole is a very strong basic compound (based on its pKa). Tioconazole is a potentially toxic compound. It was patented in 1975 and approved for medical use in 1982. It is marketed under the brand names Trosyd and Gyno-Trosyd (Pfizer, now Johnson & Johnson). A displacement reaction between 1-(2,4-dichlorophenyl)-2-(1H-imidazol-1-yl)ethanol and 2-chloro-3-(chloromethyl)thiophene is performed. Tioconazole ointments serve to treat women's vaginal yeast infections. Antimycotic imidazole derivative.

General function:
Involved in monooxygenase activity
Specific function:
Cytochromes P450 are a group of heme-thiolate monooxygenases. In liver microsomes, this enzyme is involved in an NADPH-dependent electron transport pathway. It performs a variety of oxidation reactions (e.g. caffeine 8-oxidation, omeprazole sulphoxidation, midazolam 1'-hydroxylation and midazolam 4-hydroxylation) of structurally unrelated compounds, including steroids, fatty acids, and xenobiotics. Acts as a 1,8-cineole 2-exo-monooxygenase. The enzyme also hydroxylates etoposide.
